Th17 cells are heavily involved in EAE, a mouse model of multiple sclerosis. Chung and Liao investigated the effects of knocking out CXCR3 in Th17 infiltration and glial cell cytokine production in the central nervous system (CNS).
CXCR3 is a key chemokine receptor involved in recruitment of cells to sites of inflammation. However, in Chen and Liao's work with EAE CXCR3-/- models, they actually showed increased Th17 cells in the CNS, leading to more severe EAE. When present and engaged, CXCR3 also promotes IFNγ and IL-12 production, which would antagonize Th17 development. In addition, CXCR3 signaling in glial cells was shown to suppress NF-κB, which could limit overproduction of pro-Th17 cytokines. Chen and Liao theorize that the increased Th17 cell numbers are not due to increased number of Th17 cells entering the CNS, but rather, the expansion of the infiltrating Th17 cells due to the pro-Th17 environment created by activated glial cells in the CNS parenchyma.
